Scientific Background: | Neuron-specific neurofilaments belong to the intermediate filament protein family, also known as 10nm filaments. As major constituents of the axonal cytoskeleton, neurofilaments contain heteropolymers of three major proteins: 200 kDa heavy molecular weight protein NF-H, 160 kDa medium molecular weight protein NF-M, and 68 kDa light molecular weight protein NF-L, with the ratio of each changing during neuronal development. All three proteins contain globular head and tail domains flanking a central alpha-helical domain. An accumulation of neurofilament proteins and/or mutations in one of the three major proteins occur during the progression of numerous neurological diseases, including ALS. |
